My current setup is a Plusnet Hub 2 as a router with ethernet cable linking an Asus RT-AC68U which is set to run as Access Point.

I plan to get a Grandstream HT801 and plug into the Asus access point LAN port the Asus is linked via Wan port to the Hub 2 and Hub 2 dishes out DHCP for the LAN and my PC linked to the Asus AP seems to work fine. I can't see why not for VOIP.

Access point lol a switch I would guess. There is no PPPoE or IPV6 only Wifi and LAN in the options when in AP mode.

If the Asus is acting as a switch there should be no practical difference plugging the ATA into the AP vs plugging it into the router.
